He loses his powers because of he has natural powers unlike in amazing Spider-Man he didn't have web shooters but he has natural webs out of his wrist, if he gets to stressed out he loses his ability to use powers. Him losing m.j. he gets to stressed and he needs to save her which brings him back.

I mean this is pretty clear in the film the whole time. It's shown clearly that the trauma of his life falling apart is causing his powers to falter. The movie doesn't need to explain anything, it SHOWS it. That's the beauty of film being a visual medium.
